2 Abstract Songbirds provide an ideal animal model in which to study vocal learning due to their special ability to imitate sounds they hear. Both song learning and maintenance rely on auditory memories of the target songs, and the songbird brain areas that participate in memorization of songs have been a main area of research for the last decades. A candidate brain region involved in target song memorization is the caudal medial nidopallium (NCM), a higher auditory area often compared to human auditory association cortex. Further knowledge of the NCM memory function is crucial for a deeper analysis of the mechanisms involved in vocal learning. Our main goal is to characterize the role of NCM in formation and usage of a target song memory. To this end, we combined brain lesions with different behavioral paradigms in order to link possible performance deficits with memory function. In the first project, we used the natural song learning process in juvenile birds to investigate the involvement of NCM in auditory memory. Young birds first memorize the song of an adult tutor, typically the father, and then attempt to imitate it. Existence of such a memory has been demonstrated by separating young birds from the father before they are able to imitate his song and observing that this did not prevent successful song copying. We repeated the same experiment, where in addition to removing the father we also made NCM lesions. If the targeted brain region is necessary for storing or using a memory of the tutor song, then lesions would impair song imitation. This was not the case, as we observed lesioned birds learning their tutor songs as well as control animals. Since after surgery birds had no longer access to the tutor song, we could conclude that the lesioned brain region does not contain a memory of the tutor song which is used for song learning. In a second experiment we tested whether NCM was instead necessary for memory acquisition only by making lesions before tutor song memorization. Young birds that were given exposure to a tutor song only after NCM lesions were also able to 7

3 8 ABSTRACT learn that song as well as controls did. Contrary to the first experiment, this latter one targeted not information but function, which is something that the brain may be able to compensate for. We are thus not able to rule out the involvement of NCM in tutor song memory formation. In the second project, we applied a well-established behavioral paradigm (white noise playbacks contingent to syllable pitch) to slightly yet significantly drive the songs of adult birds away from their baseline values. After the treatment is interrupted, birds spontaneously restore their original song. This transition back to baseline highlights usage of a memory of their preferred song. To investigate the role of NCM in storing or using such memory, we induced birds to change their songs and then made large bilateral NCM lesions just before letting them restore their song. While control birds succeeded in recovering their baseline song, birds with lesions showed a significant impairment. A number of control experiments allowed us to rule out hypotheses such as lesions affecting hearing or motor function. We thus concluded that the necessity of NCM in song restoration is due to a memory function. Overall, this work suggests that NCM is critically involved in storing a song memory used for adult song maintenance, but not in storing the tutor song memory in juveniles. We therefore suggest a model in which the two vocal learning processes are based on different memory mechanisms.

4 Sommario Gli uccelli canori costituiscono il modello animale ideale nel quale studiare l apprendimento vocale grazie alla loro abilità nell imitare i suoni che sentono. Sia l apprendimento che il mantenimento del canto degli uccelli si basano su memorie uditive, e le parti del cervello coinvolte nella memorizzazione di suoni sono state una principale area di ricerca nelle ultime decadi. Una regione del cervello che potrebbe avere un ruolo in memoria uditiva si chiama NCM (caudal medial nidopallium), un area uditiva secondaria comparabile alla corteccia uditiva associativa umana. Una maggiore conoscenza del ruolo di NCM nella memoria è essenziale per una ulteriore analisi dei meccanismi coinvolti nell apprendimento vocale. Il nostro principale obiettivo è di caratterizzare la funzione di NCM nell acquisizione e utilizzo di memorie uditive. A questo proposito, abbiamo abbinato lesioni cerebrali con paradigmi comportamentali al fine di collegare possibili deficit e memoria. Nel primo progetto abbiamo usato il naturale processo di apprendimento canoro in giovani uccelli per studiare la funzione di NCM nella memoria. Giovani uccelli canori memorizzano il canto di un tutor adulto, di solito il padre, e poi provano ad imitarlo. L esistenza di tale memoria è stata dimostrata separando giovani uccelli dal tutor prima che siano capaci a cantare, e osservando che questo non gli impediva di imparare una buona imitazione. Abbiamo ripetuto lo stesso esperimento, e in aggiunta alla rimozione del tutor abbiamo effettuato lesioni in NCM. Se la parte del cervello lesionata fosse stata necessaria per la memorizzazione del canto del tutor, lesioni avrebbero impedito l imitazione di quest ultimo. Al contrario, non abbiamo osservato alcuna differenza nell apprendimento in uccelli con lesioni. Dal momento che dopo le lesioni i giovani uccelli non avevano più udito il canto del tutor, possiamo concludere che la parte del cervello lesionata non contiene una memoria coinvolta nell apprendimento canoro. In un secondo esperimento abbiamo valutato se NCM è alternativamente necessario solo all acquisizione di memorie uditive facendo lesioni 9

5 10 SOMMARIO prima dell esposizione al canto di un tutor. Anche in questo caso le lesioni non hanno impedito l apprendimento. Contrariamente al primo esperimento, questo aveva come obiettivo non informazione ma una funzione. Quest ultima può essere recuperata dal cervello in caso di danneggiamento. Non siamo quindi in grado di escludere che NCM possa avere un ruolo nella memorizzazione del canto del tutor. Nel secondo progetto abbiamo applicato un paradigma comportamentale in cui possiamo indurre uccelli adulti a modificare il loro canto. Quando interrompiamo il trattamento, gli uccelli ritornano spontaneamente al loro canto iniziale. Questa transizione dimostra l utilizzo di una memoria del canto originale. Per studiare il coinvolgimento di NCM in questa memoria abbiamo indotto uccelli a modificare il loro canto e poi abbiamo eseguito lesioni in NCM prima di permettere agli uccelli di ristabilire il loro canto. Animali con lesioni hanno mostrato un chiaro deficit quando confrontati con controlli senza lesioni. Ulteriori esperimenti ci hanno permesso di escludere che il deficit sia dovuto ad un danneggiamento della funzione uditiva o motoria. Concludiamo quindi che in coinvolgimento di NCM nel recupero del canto originale è di origine mnemonica. In conclusione, questo lavoro suggerisce che NCM sia coinvolto nel memorizzare una traccia uditiva usata nel mantenimento del canto adulto, ma non nell apprendimento canoro in gioventù. Suggeriamo quindi un modello nel quale i due processi di apprendimento sono basati su differenti meccasmi mnemonici.
