The probe contains green-labeled polynucleotides (ZyGreen: excitation at 503 nm and emission at 528 nm, similar to FITC), which target sequences mapping in 12p13.2 proximal to the ETV6 gene, and orange-labeled polynucleotides (ZyOrange: excitation at 547 nm and emission at 572 nm, similar to rhodamine), which target sequences mapping in 12p13.2 distal to the ETV6 gene. Z : 0.2 ml (20 reactions of 10 µl each) The ZytoLight SPEC ETV6 Dual Color Break Apart Probe EmiNPRF is designed to be used for the detection of translocations involving the ETV6 gene at 12p13.2 in formalin-fixed, paraffinembedded tissue or cells by fluorescence in situ hybridization (FISH). The ZytoLight SPEC ETV6 Dual Color Break Apart Probe EmiNPRF must be stored at 2 8 C protected from light and is stable through the expiry date printed on the label. This product is designed for in vitro diagnostic use (according to EU directive 98/79/EC). Interpretation of results must be made within the context of the patient s clinical history with respect to further clinical and pathologic data of the patient by a qualified pathologist! 3 Do not use the reagents after the expiry date has been reached! This product contains substances (in low concentrations and volumes) that are harmful to health. Avoid any direct contact with the reagents. Take appropriate protective measures (use disposable gloves, protective glasses, and lab garments)! If reagents come into contact with skin, rinse skin immediately with copious quantities of water! A material safety data sheet is available on request for the professional user! Principle of the Method The presence of certain nucleic acid sequences in cells or tissue can be detected by in situ hybridization using labeled DNA probes. The hybridization results in duplex formation of sequences present in the test object with the labeled DNA probe. Duplex formation (with sequences of the chromosomal region 12p13.2 in the test material) is directly detected by using the tags of fluorescencelabeled polynucleotides

4 Instructions Pretreatment (dewaxing, proteolysis, post-fixation) should be carried out according to the needs of the user. Denaturation and hybridization of probe: NK Pipette 10 µl ZytoLight SPEC ETV6 Dual Color Break Apart Probe EmiNPRF each onto individual samples A gentle warming of the probe, as well as using a pipette tip which has been cut off to increase the size of the opening, can make the pipetting process easier. Avoid long exposure of the probe to light. OK Avoiding trapped bubbles, cover the samples with a coverslip (22 mm x 22 mm). Seal the coverslip, e.g. with a layer of hot glue from an adhesive pistol or with rubber cement PK= Denature the slides at 75 C (±2 C) for 10 min, e.g. on a hot plate Depending upon the age of the sample and variations in the fixation stage, it may be necessary to optimize the denaturing temperature (73 C- 77 C). QK= Transfer the slide to a humidity chamber and hybridize overnight at 37 C (e.g. in a hybridization oven) It is essential that the tissue/cell samples do not dry out during the hybridization step. Further processing, such as washing and counter-staining, can be completed according to the user s needs. For a particularly user-friendly performance, we recommend the use of a ZytoLight FISH system by ZytoVision. These systems were also used for the confirmation of appropriateness of the ZytoLight SPEC ETV6 Dual Color Break Apart Probe EmiNPRF

5 Results With the use of appropriate filter sets, the hybridization signals of the labeled chromosomal region 12p13.2 appear green and orange. In interphases of normal cells or cells without a translocation involving the 12p13.2 band, two green/orange fusion signals appear. One 12p13.2 locus affected by a translocation is indicated by one separate green signal and one separate orange signal. In order to judge the specificity of the signals, every hybridization should be accompanied by controls. We recommend using at least one control sample in which the 12p13.2 status is known. Care should be taken not to evaluate overlapping cells, in order to avoid false results, e.g. an amplification of genes. Due to decondensed chromatin, single FISH signals can appear as small signal clusters. Thus, two or three signals of the same size, separated by a distance equal to or less than the diameter of one signal, should be counted as one signal. 10 Data Sheet Principio del Metodo La presenza di determinate sequenze di acido nucleico nelle cellule o nei tessuti può essere rilevata attraverso ibridazione in situ con l'utilizzo di sonde a DNA marcate. L'ibridazione induce la formazione di una struttura a doppio filamento (ibrido duplex) tra le sequenze presenti nel campione in analisi e la sonda. La formazione del duplex (con sequenze della regione cromosomica 12p13.2) è evidenziata direttamente usando i polinucleotidi marcati con fluorofori. Istruzioni Il pretrattamento (deparaffinazione, proteolisi, post-fissazione) deve essere effettuato indipendentemente dalle esigenze dell'utilizzatore. Denaturazione e ibridazione della sonda: 1. Vortexare la sonda ZytoLight SPEC ETV6 Dual Color Break Apart Probe e dispensare 10μl per ogni campione. Distribuire a piccole gocce sull'intera superficie evitando di concentrare in un unico punto. Alternativamente, aggiungere la sonda al centro di un coprioggetto e applicare il coprioggetto sull'area in esame. Un leggero riscaldamento della sonda e l'utilizzo di un puntale tagliato in punta semplificano l'operazione. Evitare l esposizione prolungata alla luce. 2. Evitare la formazione di bolle, coprire il campione con un vetrino coprioggetto (22 mm x 22 mm). Sigillare il vetrino copri oggetto con colla a caldo o mastice. 3. Denaturare a 75 C (±2 C) per 10 minuti utilizzando una piastra calda. A seconda dell'età del campione e variazioni nella fase di fissazione, può essere necessario ottimizzare la temperatura di denaturazione (73 C-77 C). 4. Trasferire i vetrini in una camera umida e ibridare overnight a 37 C in una stufa ventilata. E importante che il campione (cellule o tessuto) non si asciughino durante l ibridazione Eventuali ulteriori processi, quali lavaggio e contro-colorazione, possono essere completati secondo le esigenze dell'utente. Per ottenere prestazioni ottimali, si consiglia l'uso di un sistema di FISH ZytoLight (ZytoVision). 11 Data Sheet Risultati: Con un appropriato set di filtri, i segnali di ibridazione con la regione cromosomica 12p13.2 appaiono verdi e arancio. In una interfase di cellule normali o in cellule senza traslocazione della banda 12p13.2, appaiono due segnali verdi/arancio fusi. In cellule con traslocazione del locus 12p13.2 è indicato dalla separazione del segnale verde e arancio. Al fine di valutare la specificità dei segnali, ogni ibridazione deve essere verificata mediante controlli (positivo e negativo). Si consiglia di utilizzare almeno un campione di controllo in cui sia inequivocabilmente nota la situazione della regione cromosomica 12p13.2. Si deve prestare attenzione a non valutare cellule sovrapposte, in modo da evitare falsi risultati, es un'amplificazione di geni. A causa decondensazione cromatina, singoli segnali FISH possono apparire come piccoli cluster di segnale. Così, due o tre segnali della stessa dimensione, separati da una distanza uguale o inferiore al diametro di un segnale, dovrebbero essere considerati come un segnale.
