3 ABSTRACT In the present study I trained common marmoset monkeys (Callithrix jacchus) to perform cognitive tasks of attention and memory using the CAmbridge Neuropsychological Test Automated Battery (CANTAB), a computerized system widely used to assess cognitive functions in humans. The CANTABallowsthe testing ofnon-human primates using the same basic proceduresemployed in human testing, thereby increasing the probability that the same cognitive functions are studied across differentprimate species. From the battery, I selected two tasks based on their relevance to human cognitive disorders, in particular Alzheimer disease: the five-choice serial reaction time (5-CSRT) task of attention, and the delayed matching-to-sample(dms) task of memory. The results demonstratedthat marmosets were able to perform 5-CSRT but not the DMS task. With regard to the latter, I developed a new memory task which could be performed by marmoset monkeys, the concurrent delayed match-to-position (CDMP) task. I pharmacologicallyvalidated the marmoset's Performance on these tasks by investigating the effects of two reference cholinergic drugs, the muscarinic reeeptor antagonist scopolamine and the nicotinic reeeptor agonist nicotine. Acetylcholine is a fundamental modulator of attention and memory, and cognitive studies in several species, including human, have demonstrated that scopolamine impairs and nicotine enhances attention- and memory-taskperformance. The results of the present study confirmed the important role of the cholinergic system in marmoset Cognition. Recently, the metabotropic glutamatergic reeeptors (mglurs) have been investigated in terms of their aetiological and pharmacotherapeutic relevance to a ränge of neuropsychiatric diseases. In particular, the high density of mglur 2 in the temporal lobe suggests that it may be implicated in memory functions. LY is a recently-developed mglur2 and 3 agonist, and whilst its effects on Cognition have not yet been widely investigated,preliminary findings in rodents indicate that it impairs memory. Therefore, I investigated the distribution of H-LY binding in the hippocampal formationof commonmarmoset monkeys and comparedthis with that in the rat. Finally, I assessed the effects of LY on marmoset Performance in the 5-CSRT and CDMP tasks. Although 3H-LY binding in marmoset brain was low compared to the rat, LY354740clearly impaired cognitive funetioning, but had no specific effect on memory. Given the low binding, future LY primate studies should be condueted in other primate species. In summary, I have demonstrated that marmosetmonkeys can be trained to perform several of the CANTABtasks and that marmoset Performance on these is sensitive to pharmacologicalchallenge of the cholinergic system. 5 RIASSUNTO Nel corso del lavoro di dottorato ho addestrato scimmie della specie Callithrix jacchus (marmoset) a svolgere un test di attenzione e un test di memoria tramite il "CAmbridge Neuropsychological Test Automated Battery" (CANTAB), sistema automatico spesso utilizzato nello studio delle funzioni cognitive dell'uomo. II CANTAB permette di esaminare le capacitä cognitive dei primati tramite una procedura analoga a quella utilizzata per esaminare le medesime capacitä negli esseri umani, aumentando pertanto l'isomorfismo proceduraletra specie umana e modello animale nell'analisi di processi di base e patologici. In base all'elevata importanzadiagnostica di questi test in malattie neuro-degenerative come il morbo di Alzheimer, ho selezionato dal CANTAB due paradigmi in grado di valutare le capacitä attenzionali e mnesiche delle callitricidi, rispettivamente il "five-choice serial reaction time (5-CSRT) test", e il "delayed matching-to-sample (DMS) test". Analisi preliminari hanno dimostrato che le marmosetriescono ad apprendere il 5-CSRT ma non il DMS: pertanto ho sviluppato un nuovo test di memoria, ü "concurrent delayed match-toposition (CDMP) test". In seguito ho sottoposto entrambi i test a validazione farmacologica studiando la Performance delle marmoset in seguito a somministrazione di nicotina e scopolamina, sostanze attive a livello del sistema colinergico. Numerosi studi condotti in animali hanno dimostrato il ruolo centrale del sistema colinergico in compiti di diverse specie memoria e attenzione. In particolare, in riferimento ai farmaci impiegati nel corso della mia tesi di dottorato, la scopolamina e risultata in grado di peggiorare, mentre la nicotina di mighorare la Performance in compiti di attenzione e memoria. I risultati ottenuti nella presente tesi confermano il ruolo centrale del sistema colinergico nella mediazione di risposte cognitive nelle scimmie marmoset. Studi recenti hanno focalizzato 1'attenzione sul ruolo esercitato dai recettori metabotropici glutamatergici (mglur) in un ampio spettro di malattie neuropsichiatriche. In particolare, l'elevata densitä dei mglur2 a livello del lobo temporalesuggerisce che questo recettore sia implicato nelle funzioni mnemoniche. II farmaco LY354740e un agonista dei mglur2 e 3 sviluppato recentemente, i cui effetti sulle funzioni cognitive non sono stati ancora adeguatamente studiati. Risultati preliminari nei roditori indicano un peggioramento in compiti di memoria in seguito alla somministrazionedi tale sostanza: pertanto ho analizzato la distribuzione di 3H-LY nella formazione ippocampale delle marmoset paragonandola a quella del ratto. Sebbene il livello di 3H-LY sia basso rispetto a quello del ratto, LY354740riduce chiaramentela Performance delle marmosetnel 5-CSRT e nel CDMP. In conclusione, ho dimostrato che le marmoset sono in grado di eseguire test

6 cognitivi tramite lo stesso strumentoutilizzato per studiare i medesimi meccanismi nell'uomo (il CANTAB) e che la loro Performance e sensibile a stimolazioni farmacologichedel sistema colinergico.pertanto, il presente lavoro rappresenta un importante passo in avanti nello studio di processi cognitivi di base e nella valutazione degli effetti di sostanze sviluppate per trattarne eventuali sviluppi patologici. 10
