ITALIANO DESCRIZIONE Mycoplasma System VET è un sistema a 24 pozzetti contenente substrati biochimici ed antibiotici essiccati per la ricerca, la determinazione semiquantitativa e l antibiogramma di Mycoplasma spp ed Ureaplasma spp. Il sistema viene inoculato con la sospensione del campione ed incubato a 36±1 C per ore. I test per la ricerca, il conteggio e l antibiogramma vengono interpretati valutando il viraggio di colore dei vari pozzetti ed eseguendo un esame microscopico. CONTENUTO DELLE CONFEZIONI 20 Sistemi Mycoplasma System VET 20 Fiale di Physiological Solution (7,0 ml) PRODOTTI NECESSARI NON CONTENUTI Olio di vaselina per uso microbiologico (ref ). PRINCIPIO DEL METODO Mycoplasma System VET permette il conteggio, l identificazione e l antibiogramma delle Mycoplasmataceae. Il sistema rende anche possibile l identificazione di Trichomonas vaginalis e Candida spp. Il conteggio delle Mycoplasmataceae viene determinato attraverso il viraggio di colore nei pozzetti 1-GR+, 2-GR++ e 3-GR+++. L identificazione di micoplasmi ed ureaplasmi si evidenzia attraverso l utilizzo di substrati biochimici presenti nei pozzetti 4-ADC e 5-UR. La presenza di Trichomonas vaginalis e Candida spp viene valutata attraverso osservazione microscopica del liquido colturale prelevato dal pozzetto 6-TR/YE. L antibiogramma viene interpretato valutando la crescita o l inibizione dei microrganismi nei pozzetti contenenti antibiotici da 7-TE a 24-AZM. RACCOLTA E CONSERVAZIONE DEI CAMPIONI I campioni devono essere inviati al laboratorio per la semina nel sistema Mycoplasma System VET subito dopo il prelievo. Evitare la conservazione in frigorifero anche per tempi limitati in quanto le basse temperature possono danneggiare la vitalità di alcuni microrganismi particolarmente sensibili come Trichomonas vaginalis pregiudicando il risultato finale. 1/8

TAMPONE Immergere il tampone (dopo il prelievo del materiale) nella fiala di Physiological Solution ed attendere 5 minuti. Strizzare con cura il tampone contro la parete della fialain modo che il materiale clinico risulti omogeneamente disperso nella soluzione fisiologica. LIQUIDI BIOLOGICI Dispensare 0.2 ml di campione nella fiala di Physiological Solution, agitare ed attendere 5 minuti prima della semina. MATERIALE DA MYCOPLASMA TRANSPORT BROTH (ref ) Dispensare 1 ml di Mycoplasma Transport Broth, contenente il campione in esame, nella fiala di Physiological Solution, agitare delicatamente ed attendere 5 minuti prima della semina. 2/8

3 INOCULO DEL SISTEMA Prelevare un sistema dal suo involucro e portarlo a temperatura ambiente. Annotare i dati relativi al campione e la data di inizio dell esame. Dispensare 0,2 ml di sospensione del campione in ciascun pozzetto del sistema. Coprire con una goccia di olio di vaselina tutti i pozzetti escluso il pozzetto 6-TR/YE. Coprire il sistema con l apposito coperchio ed incubare a 36±1 C per ore. Nel caso di sospetta presenza di Mycoplasma spp, incubare a 36±1 C per ulteriori 24 ore. Al termine dell incubazione osservare il viraggio di colore dei pozzetti ed interpretare i risultati. Ureaplasma spp: osservare il viraggio di colore del pozzetto da giallo a rosso. Trichomonas vaginalis / Candida spp: prelevare una goccia di liquido dal pozzetto, depositarla su un vetrino portaoggetto e, dopo aver deposto il vetrino coprioggetto, osservare al microscopio (40x) la presenza di protozoi ciliati mobili e/o clamidospore ed ife. Osservare il viraggio di colore dei pozzetti da giallo ad arancio-rosso. Colore giallo = assenza di crescita microbica, Sensibile. Colore arancio-rosso = crescita microbica, Resistente. 4 CONTROLLO QUALITÀ Ogni lotto di Mycoplasma System VET viene sottoposto al controllo di qualità utilizzando i seguenti microrganismi standard: Mycoplasma hominis ATCC 23114, Ureaplasma urealiticum ATCC 27618, Candida albicans ATCC FATTORI CHE POSSONO INVALIDARE I RISULTATI Imprecisa standardizzazione dell inoculo; materiale da esaminare inadatto; uso di sistemi e reagenti supplementari scaduti; temperatura e tempi di incubazione non rispettati. PRECAUZIONI Il prodotto Mycoplasma System VET non contiene sostanze nocive in concentrazioni superiori ai limiti fissati dalla normativa vigente, perciò non è classificato come pericoloso; per il suo impiego si consiglia comunque di consultare la scheda di sicurezza. Mycoplasma System VET è un dispositivo monouso da utilizzare in un ambito professionale, in laboratorio e da operatori adeguatamente addestrati, con metodi approvati di asepsi e di sicurezza nei confronti degli agenti patogeni. CONSERVAZIONE Conservare Mycoplasma System VET a 2-8 C nella sua confezione originale. Non conservare vicino a fonti di calore ed evitare eccessive variazioni di temperatura. In queste condizioni il prodotto è valido fino alla data di scadenza indicata in etichetta. Non utilizzare oltre questa data. Eliminare se vi sono segni di deterioramento. ELIMINAZIONE DEL MATERIALE UTILIZZATO Dopo l utilizzazione, Mycoplasma System VET e il materiale venuto a contatto con il campione devono essere decontaminati e smaltiti in accordo con le tecniche in uso in laboratorio per la decontaminazione e lo smaltimento di materiale potenzialmente infetto. 5 Mycoplasma System VET System for count, identification and susceptibility testing of mycoplasmas in the veterinary field. ENGLISH DESCRIPTION Mycoplasma System VET is a 24-well system containing desiccated biochemical and antibiotic substrates for the detection, semi-quantitative count and susceptibility testing of Mycoplasma spp and Ureaplasma spp. The system is inoculated with the sample suspension and incubated at 36±1 C for hours. The tests results are interpreted by assessing the color change in the various wells and with microscope examination. CONTENT OF THE KIT 20 Mycoplasma System VET 20 Vials of Physiological Solution (7.0 ml) ITEMS NECESSARY BUT NOT INCLUDED IN THE KIT Vaseline oil for microbiological use (ref ). PRINCIPLE OF THE METHOD Mycoplasma System VET allows count, identification and susceptibility testing of Mycoplasmataceae. The system also provides the detection of Trichomonas vaginalis and Candida spp. The count of Mycoplasmataceae is evaluated by the color change in the wells 1-GR+, 2-GR++ and 3-GR+++. Identification of mycoplasmas and ureaplasmas is obtained with biochemical reactions in the wells 4-ADC and 5-UR. The presence of Trichomonas vaginalis and Candida spp is determined by microscope examination of a drop of culture liquid taken from well 6-TR/YE. Susceptibility testing is assessed on the basis of the growth or inhibition of the microorganisms in the wells 7-TE to 24-AZM containing antibiotics. COLLECTION AND CONSERVATION OF THE SAMPLES The samples must be sent for inoculation in the Mycoplasma System VET immediately after they are taken. Do not store in the refrigerator for even limited periods of time, since low temperatures may damage the vitality of some particularly sensitive microorganisms such as Trichomonas vaginalis prejudicing the final result. 5/8

SWAB Immerse the swab (after obtaining the clinical material) in the vial of physiological solution and wait 5 minutes. Carefully squeeze the swab against the vial wall so that the clinical material is dispersed homogeneously in the Physiological Solution. BIOLOGICAL FLUIDS Add 0.2 ml of sample to the vial of Physiological Solution, shake gently and wait 5 minutes before inoculating. SAMPLE MATERIAL FROM MYCOPLASMA TRANSPORT BROTH (ref ) Add 1 ml of Mycoplasma Transport Broth, containing the sample under examination, to the vial of Physiological Solution, shake gently and wait 5 minutes before inoculating. 6/8

7 INOCULATION OF THE SYSTEM Take a system from its wrap and bring it to room temperature. Write down the name of the patient and the date of the start of the examination. Dispense 0.2 ml of sample suspension into each well of the system. Overlay with 1 drop vaseline oil all wells other than well 6-TR/YE. Cover the system with the lid provided and incubate at 36±1 C for hours. If the presence of Mycoplasma spp is suspected, incubate at 36±1 C for further 24 hours. After incubation watch for the color change in the wells and interpret the results. Trichomonas vaginalis / Candida spp: Take a drop of liquid from the well, deposit it on a glass slide, place a cover slip on top and examine at the microscope (40x) for the presence of mobile ciliated trophozoite and/or chlamydospores and hyphae. 8 QUALITY CONTROL Each batch of Mycoplasma System VET is subjected to quality control using the following reference strains: Mycoplasma hominis ATCC 23114, Ureaplasma urealiticum ATCC 27618, Candida albicans ATCC FACTORS THAT MAY INVALIDATE THE RESULTS Poor standardization of the inoculm; unsuitable material to examine; use of expired systems or expired supplementary reagents; non compliance with temperatures and/or times of incubation. PRECAUTIONS The product Mycoplasma System VET does not contain hazardous substances in concentrations exceeding the limits set by current legislation and therefore is not classified as dangerous. It is nevertheless recommended to consult the safety data sheet for its correct use. Mycoplasma System VET is a disposable device to be used for in vitro diagnostic use only. It is intended for professional use by properly trained personnel in the laboratory, using approved asepsis and safety methods for handling pathogenic. CONSERVATION Store Mycoplasma System VET a at 2-8 C in the original packaging. Keep away from sources of heat and avoid excessive changes in temperature. In such conditions the product will remain valid until the expiry date indicated on the label. Do not use beyond that date. Eliminate without using if there are signs of deterioration. DISPOSAL OF USED MATERIAL After use, Mycoplasma System VET and material that has come into contact with the sample must be decontaminated and disposed of in accordance with the techniques used in the laboratory for decontamination and disposal of potentially infected material.
