ITALIANO DSCRIZION NTROSYSTM 18R è un sistema a 18 pozzetti contenenti substrati biochimici per l identificazione degli enterobatteri Gram negativi, ossidasi negativi. Il sistema viene inoculato con la sospensione batterica del microrganismo in esame ed incubato a 36 ± 1 C per ore. Il microrganismo viene identificato mediante codifica numerica ottenuta dai viraggi cromatici dei vari test biochimici. L'identificazione si basa su prove biochimiche eseguite su terreni colturali contenenti substrati specifici presenti nei pozzetti. La combinazione delle reazioni positive e negative permette la formazione di un codice numerico che consente a sua volta di identificare il batterio in esame, con l'aiuto dell' NTROSYSTM 18R COD BOOK (cod ) e dell' IDNTIFICATION COD DISK (cod ). 1

PROCDURA DL TST PRPARAZION DLLA SOSPNSION BATTRICA 1. Il microrganismo da identificare deve essere di isolamento recente (18-24 ore); batteri provenienti da colture con più di 48 ore possono dar luogo a risultati non attendibili. 2. Prima di procedere alla semina del microrganismo in esame, è necessario eseguire la colorazione di Gram ed il test dell' ossidasi sullo stesso. Solo batteri gram negativi, ossidasi negativi possono essere seminati su NTROSYSTM 18R. Per la corretta esecuzione di entrambi i test si rimanda ai manuali di batteriologia idonei. 3. Aprire una fiala di soluzione fisiologica sterile a ph controllato contenuta nel kit. 4. Prelevare una o più colonie morfologicamente simili, ben isolate, dal terreno di coltura solido e sospenderle nella soluzione fisiologica in modo da ottenere una torbidità equivalente a 0,5 McFarland (Sospensione batterica). 5. Omogeneizzare accuratamente. INOCULO DL SISTMA 1. Prelevare un sistema dal suo involucro e portare a temperatura ambiente. 2. Annotare data e provenienza del germe. 3. Trasferire 0.2 ml (4 gocce) di Sospensione batterica in ciascun pozzetto del sistema e coprire con 2 gocce di olio di vaselina per uso microbiologico i pozzetti 2-LDC, 3-ODC, 4-ADC, 7-UR e 8-H 2S. 4. Coprire il sistema con l apposito coperchio ed incubare a 36 1 C per ore. INTRPRTAZION DI RISULTATI Al termine dell incubazione: 1. Introdurre 2 gocce di alfa-naftolo ed 1 goccia di NaOH 40% nel pozzetto 10-VP. Attendere minuti ed osservare la comparsa di una colorazione rosa-rossa. 2. Introdurre 2 o 3 gocce di Kovac s Reagent (cod ) nel pozzetto 11-IND. Attendere 1 o 2 minuti ed osservare la per la comparsa di una colorazione rossa. 3. Osservare il viraggio di colore dei pozzetti ed interpretare i risultati servendosi della tabella n Trascrivere il risultato sul modulo raccolta dati e formare il codice numerico di 6 cifre seguendo le istruzioni riportate nel paragrafo FORMAZION DL CODIC NUMRICO. 5. Risalire quindi all identificazione batterica servendosi dell' NTROSYSTM 18R COD BOOK (cod ) o dell' IDNTIFICATION COD DISK (cod ). 2

NGLISH DSCRIPTION NTROSYSTM 18R is a 18 wells system containing desiccated biochemical substrata for the identification of Gram-negative, oxidasenegative enterobacteria. The system is inoculated with the suspension of the microorganism to test and incubated at 36 ± 1 C for hours. The microorganism is identified by evaluating the color change in the different culture media in the wells and by a code number obtained from biochemical reactions interpretation. TST PROCDUR PRPARATION OF TH BACTRIAL SUSPNSION 1. The microorganism to identify must have been recently isolated (18-24 hours); bacteria from more than 48 hours incubation can provide not reliable results. 2. Before inoculating the microorganism to test, it is necessary to perform Gram staining test and oxidase test. Only Gram-negative, oxidase-negative bacteria can be inoculated on NTROSYSTM 18R. Consult suitable manuals of bacteriology to correctly perform these tests. 3. Open one vial of sterile physiological solution contained in the kit. 4. Take one or more morphologically similar, well-isolated colonies from the agar medium and suspend in the physiological solution so to obtain a turbidity equivalent to 0,5 McFarland (Bacterial suspension). 5. Thoroughly homogenize. INOCULUM OF TH SYSTM 1. Take a system from its pouch and leave it at room temperature. 2. Note date and origin of the microorganism. 3. Transfer 0.2 ml (4 drops) of Bacterial suspension into each well of the system and cover the wells 2-LDC, 3-ODC, 4-ADC, 7-UR and 8-H 2S with 2 drops of vaseline oil. 4. Cover the system with the lid provided and incubate at 36 ± 1 C for hours. 6

9 INTRPRTATION OF RSULTS 1. Subsequently to the incubation: 2. Introduce 2 drops of alpha-naphtol and 1 drop of NaOH 40% in the well 10-VP. Wait minutes and watch for the appearance of a pink-red color. 3. Introduce 2 or 3 drops of Kovac s Reagent (ref ) in the well 11-IND. Wait 1-2 minutes and watch for the appearance of a red color. 4. Observe the color change of the wells and interpret the results using the table no Note the result on the Test Results Form and build up the 6-digit code following the instructions provided in the paragraph COD NUMBR FORMING. 6. Identify the microorganism using the NTROSYSTM 18R COD BOOK (ref ) or the IDNTIFICATION COD DISK (ref ). CAUTIONS The product, NTROSYSTM 18R, is not classified as hazardous under current legislation, however refer to the safety data sheet for a correct use. NTROSYSTM 18R is a disposable device to be used only for in vitro diagnostic use. It must be used in the laboratory by properly trained personnel, using approved aseptic and safety methods for handling pathogenic agents. STORAG Store NTROSYSTM 18R at 2-8 C in its original container. Keep away from sources of heat and avoid excessive changes of temperature. In such conditions NTROSYSTM 18R maintains its validity until the expiry date indicated on the label. Do not use beyond that date. liminate without using if there are signs of deterioration.
