4 1 Introduzione Il kit QuickToxTM Kit for QuickScan detection of Ochratoxin è stato validato su vino. Il kit QuickTox Ochratoxin A Quantitative kit for Wine fornisce risultati quantitativi di contaminazione di ocratossina A nel range ppb. 2 Contenuto del kit 50 QuickTox Stips* Buffer DB3 Extraction Buffer for Wine (Cat. #OTA-EBW)** 100 puntali * Le strip sono sensibili all umidità pertanto si consiglia di conservarle all interno del tubo ben chiuso. Evitare di piegare le strips in quanto questo ne comprometterebbe il normale funzionamento. ** Leggere attentamente la scheda di sicurezza dell Extraction Buffer for Wine (Cat. #OTA-EBW). 3 Preparazione del test 1. Accendere il termoblocco e settarlo a 22 C. Attendere almeno 10 minuti prima dell esecuzione del test. Assicurarsi che la temperatura sia stabile e che lo schermo indichi OK. 2. Lasciare che campioni e reagenti raggiungano la temperatura ambiente (20-24 C) prima di iniziare i test. 4 Preparazione del campione 1. Prelevare, dopo aver agitato vigorosamente, un campione rappresentativo in accordo con il suo piano di campionamento. È comunque raccomandato prelevare almeno 750 ml di campione. 2. Degassamento (per campioni di vino frizzante): Trasferire 20 ml di campione in un tubo da centrifuga da 50 ml e agitare vigorosamente a mano per 30 secondi; aprire il tubo per rimuovere biossido di carbonio. Ripetere 3 volte. 3. Trasferire 5 ml di vino in un tubo da centrifuga da 15ml o da 50ml. Nel puntale della pipetta non devono essere presenti bolle; in caso contrario ripetere lo step di degassamento. 4. Agitare per 20 secondi l Extraction Buffer for Wine. 5. Aggiungere 2 ml di Extraction Buffer for Wine nel tubo da centrifuga contenente il campione di vino. 6. Chiudere il tubo e agitare vigorosamente a mano per 30 secondi. 7. Filtrare il campione con carta da filtro o, in alternativa, centrifugare per 5 minuti a 2000 g (utilizzare il surnatante per la fase successiva). 5 - Esecuzione del test 1. Trasferire 50 µl di Buffer DB3 in un tubo di reazione 2. Aggiungere 200 µl di campione 3. Aspirare e ri-dispensare il campione con una pipetta per 4-5 volte per miscelare 4. Inserire il tubo di reazione nell incubatore settato a 22 C (accertarsi che l incubatore abbia raggiunto la temperatura di 22 C) 5. Far acclimatare il campione per 2 minuti a 22 C nell incubatore 6. Inserire immediatamente una strip nel campione con la freccia rivolta verso il basso 7. Attendere 10 minuti 8. Rimuovere la strip dal campione 9. 5 6 Lettura della strip con il QuickScan Nota: per la lettura il software QuickScan deve essere in modalità di lettura dei segnali. In caso di dubbi, contattare Generon S.p.A. 1. Fare doppio clic sull icona QuickScan (QS) per aprire il programma di lettura. Apparirà il menu principale (Fig1). 2. Inserire la strip nel carrello con il codice a barre rivolto verso il basso e vicino all impugnatura (Fig.2). 3. Inserire Il carrello nel lettore (Fig.3). 4. Leggere la strip cliccando su "Read Test" sul software QS e attendere che il QS termini la scansione. 5. Nella finestra dei risultati (Fig. 4) è possibile inserire i dettagli identificativi dell analisi. Affinché l immagine venga salvata, inserire almeno il nome del campione nello spazio Sample ID e premere Invio (Enter) sulla tastiera. Fig. 1 Fig. 2 Fig. 3 Fig Se si desidera stampare immediatamente il report di analisi, premere il comando PRINT in basso a destra. 7. Premere CLOSE per chiudere la finestra di lettura. 8. Per rivedere o stampare i risultati in un secondo momento, doppio clic sulla cartella REPORTS sul desktop. 9. Per visualizzare il registro di tutte le analisi eseguite, cliccare sul comando DATA LOG nella schermata iniziale del programma Quick Scan. 7 Calcolo dei risultati Per il calcolo dei risultati usare il foglio di calcolo "Spreadsheet_GENERON_OchratoxinA_Wine". 1. Inserire manualmente i valori REF indicati nel Certificato di Qualità (fornito nel kit) nelle celle arancioni. Questi valori sono lottospecifici e non possono, quindi, essere utilizzati con lotti diversi. 2. Inserire manualmente i valori SI ottenuti dalla lettura dei campioni nelle celle gialle. 3. I risultati in ppb vengono calcolati automaticamente nella colonna Risultati. Inserire I valori REF indicati nel CQ Inserire I valori SI dei campioni Vedere i risultati del test I risultati sono riportati nell intervallo da 0.4 a 3.0 ppb. 8 1 - Introduction QuickToxTM Kit for QuickScan detection of Ochratoxin was validated for wine. QuickTox Ochratoxin A Quantitative kit for Wine provides quantitative results of ochratoxin A contamination in the range ppb. 2 Kit Components 50 QuickTox Stips* DB3 Buffer Extraction Buffer for Wine (Cat. #OTA-EBW)** 100 pipette tips * Strips are sensitive to humidity, so it is advisable to keep them inside the tightly closed tube. Avoid bending of strips as this would compromise normal running of the sample. ** Carefully read safety informations of Extraction Buffer for Wine (Cat. #OTA-EBW). 3 Test preparation 1. Switch on the incubator and set it at 22 C. Wait at least 10 minutes before starting tests. Make sure that temperature is stable and that the display shows OK. 2. Make sure that samples and reagents are at room temperature (20-24 C) before you start with tests. 4 Sample preparation 1. Collect a representative sample according to your own sampling plan after a vigorous agitation. In any case, it is recommended to take at least 750 ml of sample. 2. Degassing (for sparkling wine): Trasfer 20 ml of sample in a 50 ml tube and vigorously shake it by hand for 30 seconds; open the tube to remove carbon dioxide. Repeat 3 times. 3. Trasfer 5 ml of wine in a 15 ml or 50 ml centrifuge tube. You should not have bubbles in the pipette tip; if there are babbles in the tip it is necessary to repeat the degassing step. 4. Shake the Extraction Buffer for Wine for 30 seconds. 5. Add 2 ml of Extraction Buffer for Wine in the tube containing wine sample. 6. Cap sample and vigorously shake by hand for 30 seconds. 7. Filter the sample with qualitative filter paper or centrifuge for 5 minutes at 2000 g (use the supernatant for the following step). 5 Test Procedure 1. Trasfer 50 µl of DB3 Buffer in a reaction tube 2. Add 200 µl of sample 3. Mix well by pipetting up and down 4-5 times 4. Insert the reaction tube in the incubator set at 22 C (make sure the incubator reached the correct temperature) 5. Allow the sample to acclimate for 2 minutes at 22 C in the incubator 6. Immediately insert a strip into the sample with the arrow pointing into the reaction tube 7. Wait 10 minutes 8. Remove the strip 9. Immediately cut off and discard the bottom section of the strip covered by the arrow tape 10. 9 6 Use of the QuickScan System To perform this test you must have the QuickScan software in raw signals mode. If you have any doubt, please contact Generon S.p.A. 1. Double click on QuickScan (QS) icon to open the software. The main menu will appear on the screen (Fig1). 2. Insert the strip into the carrier with barcode facing down and near to the handle (Fig.2). 3. Insert the carrier into the reader (Fig.3). 4. Read the strip by clicking on "Read Test" and wait until the end of the scanning. 5. In result window (Fig. 4) it is possible to insert identification details of the analysis. In order to save the image, insert at least the sample ID and press Enter on the keyboard. Fig. 1 Fig. 2 Fig. 3 Fig If you want immediately print analysis report, click PRINT. 7. Click CLOSE to close the reader windows. 8. In order to see or print results in a different moment, double click REPORTS folder on the desktop. 9. In order to get access to the page with all the performed analysis, click DATA LOG on the main page of Quick Scan software. 7 Result calculation In order to calculate results you need to use the " Spreadsheet_GENERON_OchratoxinA_Wine" spreadsheet. 1. Manually insert REF values provided in the Quality Certificate (supplied with the kit) in the orange cells. These values are lot-specific and can NOT be used with different kit lots. 2. Manually insert SI values obtained with the scanning of the strips in yellow cells. 3. Results will be automatically calculated and are expressed in ppb in the Results column. Input the REF values provided in the QC Input the SI value of sample View test results Results are reported in the range ppb.
