PRINCIPIO Il triptone è il prodotto dell idrolisi enzimatica della caseina e contiene una miscela di peptide ed amminoacidi liberi. Il glucosio è una risorsa energetica. L estratto di lievito è una fonte di amminoacidi liberi e vitamine del gruppo B. L agar è l agente solidificante. TECNICA Aprire il blister e prelevare la piastra RODAC. Rimuovere il coperchio della confezione e premere delicatamente la superficie dell agar sulla superficie da esaminare per almeno 10 secondi. Altrimenti la piastra può essere utilizzata per l analisi dell aria, campionando, se possibile, 100-500 l d aria per mezzo di un campionatore SAS. Ricoprire la piastra con il coperchio ed incubare:! a 25-30 C per 68 ± 2 h per batteri mesofili;! a 25 ± 1 C per almeno 7 giorni (controllare la crescita dal secondo giorno in poi) per lieviti e funghi;! a 30 ± 1 C per 68 ± 4 h, lontano da fonti luminose, per i batteri ambientali. INTERPRETAZIONE DEI RISULTATI Osservare la formazione di Unità Formanti Colonie (UFC) ed effettuare il conteggio in relazione al volume di campione analizzato. CONDIZIONI DI CONSERVAZIONE E TRASPORTO Il prodotto può essere conservato a 10-25 C al riparo dalla luce, fino alla data di scadenza indicata in etichetta. La conservazione o il trasporto a 2-10 C non alterano in nessun modo l'efficienza del prodotto. Eliminare se vi sono segni evidenti di deterioramento o contaminazione. AVVERTENZE E PRECAUZIONI Il prodotto non contiene sostanze nocive in concentrazioni superiori ai limiti fissati dalla normativa vigente, perciò non è classificato come pericoloso; per il suo impiego si consiglia comunque di consultare la scheda di sicurezza. Il prodotto è destinato esclusivamente ad un uso professionale e deve essere utilizzato da parte di personale qualificato. TECHNICAL SHEET Page 1 of 2 Medium for surfaces and air total bacterial count TYPICAL FORMULA (g/l) Tryptone 5.0 Glucose 1.0 Yeast Extract 2.5 Agar 15.0 Final ph 7.0 ± 0.2 DESCRIPTION is a medium suggested by APHA, AOAC, ICMSF, ISO for surfaces and air total count of aerobic bacteria, prepared according to the formulation reported by ISO 4833. PRINCIPLE Tryptone is a product obtained by a controlled enzymatic hydrolysis of casein and contains a mix of peptides and free amino acids. Glucose is a source of energy. Yeast extract is a source of amino acids and vitamins of group B. Agar is the solidifying agent. Remove the first wrapper of the packet and the inner one. Remove the cap of the plate and gently push the agar surface on a level surface to examine for at least 10 seconds. Otherwise the plate can be used for air analysis, sampling, if it is possible, 100-500 l of air with an air sampler. Cover the plate with the cap. Incubate:! at 25-30 C for 68±2 h for mesophilic bacteria;! at 25 ± 1 C for minimum 7 days (from 2nd day on control for growth) for yeasts and fungi;! at 30 ± 1 C for 68 ± 4 h in the dark for environmental bacteria. INTERPRETATION OF RESULTS Observe for the formation of Units Forming Colonies (UFC) and count them referring to the volume of sample analysed. STORAGE AND TRANSPORT CONDITIONS 10-25 C away from light, until the expiry date on the label. Storage or transport at 2-10 C do not alter in any way the performance of the product. Eliminate if signs of deterioration or contamination are evident. WARNING AND PRECAUTIONS The product does not contain hazardous substances in concentrations exceeding the limits set by current legislation and therefore is not classified as dangerous. It is nevertheless recommended to consult the safety data sheet for its correct use. The product is designed for professional use and must be used only by properly trained operators. PRINCIPE La tryptone est obtenue par hydrolyse enzymatique contrôlée de la caséine et contient un mélange de peptides et d acides aminés libres. Le glucose représente la source énergétique. L extrait de levure est une source d aminoacides et de vitamines B. L agar est un agent gélifiant. Enlever le blister individuel. Enlever le couvercle supérieur et appuyer légèrement la surface gélosée contre la surface à contrôler, maintenir la pression pendant au moins 10 secondes. Il est aussi possible de prélever 100-500 litres d air lors d un contrôle d ambiance. Remettre le couvercle sur la boite. Incuber :! 68 ±2 h à 25-30 C pour la flore mésophyle aérobique,! Minimum 7 jours (avec un contrôle journalier) à 25 ± 1 C pour les levures et moisissures,! 68 ± 4 h dans l obscurité à 30 ± 1 C pour les tests environnementaux. INTERPRÉTATION DES RÉSULTATS Observer la formation d Unité Formant Colonie (UFC) et les comptabiliser en se référant à la surface ou au volume d aire contrôlé. CONDITIONS DE STOCKAGE ET DE TRANSPORT 10-25 C à l abri de la lumière jusqu à la date de péremption indiquée. Le stockage ou le transport à 2-10 C ne modifie en rien la performance du produit. Éliminer si les signes de détérioration ou de contamination sont évidents. MISE EN GARDE ET PRÉCAUTIONS Le produit ne contient pas de substances dangereuses dans des concentrations dépassant les limites fixées par la législation en vigueur et n'est donc pas classé comme dangereux.
