4 The commonly accepted view of a "fixed, ended, immutable" (Ramón y Cajal et al. 1991) central nervous system (CNS) has evolved considerably over the past few decades. The adult CNS is now considered a tissue in which adaptive events are continuously and spontaneously occurring throughout the life of an individual, either in health or after trauma. These adaptive changes are frequently referred to as neuroplasticity, defined as gain of novel properties by a system having reorganized at least part of its structural characteristics as a result of the transient action of internal (e.g., physiological) or external (environmental) constraints (Will et al. 2008). In vitro and in vivo studies reveal that the main anatomical substrates of such plasticity are axons and dendritic spines, whose dynamics result in changes of neuronal properties, connectivity and synaptic strength. Although inherited, the plastic potential of the adult CNS is very limited compared to the juvenile CNS, in which a critical period of intense plasticity has been described. Several factors restrict plasticity in the adult CNS. Highly inhibitory extracellular cues have been identified in the mature extracellular matrix (ECM) in which neurons are embedded. A key inhibitory role is played by chondroitin sulfate proteoglycans (CSPGs), the most abundant components of the mature ECM. These proteins are diffusely expressed in the extracellular space or aggregated as perineuronal nets (PNNs) mainly around parvalbumin expressing interneurons. Enzymatic digestion of CSPGs with the bacterial enzyme chondroitinase ABC (ChABC) restores plasticity in the adult CNS and promotes functional recovery after injury. How CSPGs restrict plasticity and how ChABC-mediated CSPG digestion enhances plasticity is still elusive and needs to be further investigated. The aim and novelty of my scientific endeavour was to investigate the fine and dynamic interplay between the mature ECM and CNS plasticity, by means of both in vitro and in vivo models. In particular, my studies focused on discerning the role of diffused CSPGs and PNNs in restricting structural and functional remodelling of mature neuronal circuits. Chapters I describes the establishment and characterization of in vitro cortical organotypic slice cultures to study the effects of ChABC-mediated CSPG digestion on dendritic spine 8

5 dynamics. Although we show that this preparation is suitable to study and interfere with the mature ECM, the same system lacks the reliability and reproducibility for live imaging of spine dynamics. The use of an alternative slice preparation, organotypic slice cultures of hippocampus, was therefore chosen. Chapter II describes how this preparation, in combination with imaging and electrophysiological techniques, allowed us to reveal that ChABC-mediated CSPG digestion significantly enhances the dynamic remodelling of dendritic spines. This effect is β1-integrin dependent but independent of PNN degradation. These observations identify perisynaptic CSPGs as key extracellular matrix molecules actively restricting spine dynamics and suggest a new mechanism by which ChABC treatment might promote functional plasticity. Finally, my doctoral studies focused on investigating the role of cortical PNNs in the spontaneous structural and functional rewiring of the corticospinal tract (CST) after spinal lesion. In Chapter III, we demonstrate for the first time that a complete thoracic transection of the CST induces complex and discrete spatial changes in the expression of GABAergic inhibitory markers and PNNs in the mouse sensorimotor cortex. Promoting digestion of cortical PNNs by local intracortical ChABC injection prevents CST sprouting and transiently worsens fine motor control deficits after CST transection. Thus, we identify cortical PNNs as key regulators of the CST functional remodelling after lesion. Two short Appendices follow the three main Chapters. Appendix 1 describes the attempt to establish a new transgenic mouse model to study the role of the receptor TrkB in CST homeostasis and response to injury. Lastly, Appendix 2 presents my contribution to the characterization of the synaptic functions of calsyntenin-1 in acute slices from functional calsyntenin-1 knockout and wild type mice. 9

7 Grazie al lavoro di brillanti neuroscienziati, il dogma sull immutabilità del cervello adulto sostenuto da Ramon y Cajal ( ), grande neurofisiologo scopritore dei principi base dei processi neurali, è ormai da tempo superato. Evidenze empiriche mostrano il cervello umano come un organo in grado di modificarsi spontaneamente in risposta a stimoli interni o esterni. Il termine neuroplasticità descrive la riorganizzazione strutturale e funzionale dei circuiti neurali. Tale riorganizzazione è basata sul rimodellamento delle connessioni neurali a livello di assoni e spine dendritiche, il cui dinamismo consente la formazione di circuiti alternativi che meglio si adattano agli stimoli ambientali, o la ricostruzione anatomica e funzionale dei circuiti stessi una volta danneggiati. Benché presente durante l intero arco di vita dei mammiferi, la neuroplasticità del sistema nervoso centrale è molto limitata nell adulto rispetto ai primi anni di vita. Questi ultimi sono caratterizzati da un periodo critico di intensa riorganizzazione dei circuiti neurali in risposta a stimoli ambientali. Molteplici fattori limitano la neuroplasticità del sistema nervoso centrale adulto. Tra questi, alcuni componenti della matrice extracellulare, in cui sono immersi i neuroni, possiedono proprietà che inibiscono fortemente il rimodellamento dei circuiti neurali. Un ruolo di primaria importanza è giocato dai proteoglicani condroitin solfati, i componenti più abbondanti della matrice extracellulare del sistema nervoso centrale adulto. I proteoglicani condroitin solfati sono sia secreti nella matrice extracellulare sia condensati nelle reti perineuronali che circondano una specifica classe di interneuroni. La digestione dei proteoglicani condroitin solfati e delle reti perineuronali tramite l enzima batterico condroitinasi ABC potenzia la neuroplasticità adulta e favorisce il recupero funzionale dopo una lesione del sistema nervoso centrale. I meccanismi tramite cui i proteoglicani condroitin solfati inibiscono il rimodellamento dei circuiti neurali e le dinamiche con cui l enzima batterico condroitinasi ABC potenzia la neuroplasticità adulta sono ancora oggetto di intensi studi. L obiettivo del mio lavoro è stato quello di studiare la reciproca e dinamica interazione tra la matrice extracellulare e la neuroplasticità tramite l utilizzo di modelli in vivo e in vitro. In 11

8 particolare, il mio studio si è focalizzato sul comprendere e descrivere il ruolo dei proteoglicani condroitin solfati nella restrizione strutturale e funzionale della neuroplasticità. Il Capitolo I descrive lo sviluppo di un modello in vitro (colture organotipiche di corteccia murina) finalizzato a tale scopo. Questo tipo di preparazione si è rivelato adatto allo studio della matrice extracellulare ma non della neuroplasticità strutturale. L uso di colture organotipiche di ippocampo murino, in combinazione con tecniche di microscopia in vitro e elettrofisiologia, ci ha permesso di identificare il ruolo chiave dei proteoglicani condroitin solfati perisinaptici nella restrizione della plasticità delle spine dendritiche e di descriverne i meccanismi sottostanti. Su questo studio è incentrato il Capitolo II. Il terzo Capitolo è rivolto allo studio degli effetti di una lesione alla spina dorsale sulle reti perineuronali corticali murine. Questo lavoro dimostra come la completa recisione del tratto corticospinale determina cambiamenti nella struttura delle reti perineuronali nella corteccia motoria. Inaspettatamente, la digestione di tali reti a livello corticale tramite l enzima condroitinasi ABC induce un transitorio peggioramento delle funzioni motorie e ostacola la neuroplasticità strutturale assonale che spontaneamente si manifesta nella spina dorsale in risposta alla recisione. Questi dati supportano l esistenza di una reciproca interazione tra la matrice extracellulare e la neuroplasticità dei circuiti neurali. Due Appendici seguono i tre capitoli principali. L Appendice 1 descrive lo sviluppo di un nuovo modello animale transgenico finalizzato allo studio del ruolo del recettore TrkB nell omeostasi del tratto corticospinale e nella sua risposta ad una lesione. L Appendice 2 descrive il mio contributo nella caratterizzazione delle funzioni sinaptiche della proteina calsyntenin-1. 12
